Hi All,

i have encountered a rather strange problem.

I recently performed the yeast-2-hybrid experiment with a cDNA library to identify potential interacting partners to my target protein. After performing the experiment and plating cells on media with X-gal, i do see Blue colonies, but...

rather than growing on the agar, the colonies seem to enter the agar and grow into it. I tried to isolate plasmid from these colonies, but was not succesful.

Any clues as to why they may be growing into the agar?
