Expressing protein !! Help!

Hi everyone!

I have a big problem with expressing protein
When i ligated my gene on vector successfully,
and tried many different conditions
My gene didn't OVERexpress!!

Now I want to use primer including my gene's promoter
And add some transcription factor to try to express protein

Does it work! :shock:
Give me some suggestion please!!

Ya; it should be.........

First you should have transfered them in first experiment...... as its comon that if you are inserting a new gene then promotor should be there with it if you need results fast.......

one thing that may be worth looking at is the codon usage, see if u have many rare codons.
