RNA recovery from cell cultured in agarose matrix

Post by ppm58 » Thu Mar 17, 2016 4:45 pm

I culture cells as 3D model (spheroid) embedded in agarose matrix. I am trying to recovery RNA from these culture to perform RT PCR but I meet several problem as RNA quantity (<30 ng/mL) and as RNA quality (260/280/230 ratio are bad).

Have some ideas to obtain a fair RNA quality and quantity in these culture agarose conditions.

Thank you for your help

This is a general question. For detail, how do you extract RNA? The most important thing is separating your cells and agarose, how did you do?
