peroxidase screening for actinomycetes

Post by fishbonedoris » Wed Jan 10, 2007 8:20 am

I'm currently doing my thesis on screening for enzymes in actinomycetes. One of it is to screen for peroxidase enzyme. My supervisor gave me a short method as follows:

Peroxidase test

Incubate actinomycete culture on oat meal agar for 10 days in dark at 27 oC.
Make wells of 1.0 cm in diameter.
Drop one drop of hydrogen peroxide following by one drop of pyrogallic acid.
Leave it for 15 – 30 minutes to see the result.
Use 95% ethanol as control.

But i can't seem to find any article citing this method as i need to know the concentration needed. Pls help.
