Protoplast Isolation - first step - enzyme digestion

Post by bravebeaker » Fri Dec 16, 2011 9:02 am

Hello all,
I have been reading lots of papers about protoplast isolation. I found that most of the papers but not all say that enzyme digestion is in the dark. Why should it be in the dark? i have been trying to get an explanation but cant find any. In the mean time I am trying to enzyme digestion with and without light and see the difference but whats the purpose of digestion in dark conditions from 2 to 3 hrs?

AHH, Gravity! Such a cold cruel mistress.
