Almost all proteins have secondary structure of some kind--even globular proteins. If you are asking how to tell what kind of structures are there, then you are talking about things like ORD-CD (tells you something about the relative amount of helix vs strands), spectroscopy (noes from nmr tells you something about helices and strands), centrifugation (tells you something about the overall shape). You can always attempt to crystallize a protein and determine its x-ray structure. That way you can actually "see" the secondary, tertiary, and sometimes quaternary structures of a protein or protein complex.
