so you use SalI in the ligation reaction and now you want to remove the insert again with SalI... is this what you're doing? (I didn't get it quite well, sorry) If this is the case, it's very usual, because sometimes, when you ligate 2 fragments, the restriction site is altered and isn't recognized ...

like seaseasea mentioned, if you don't have at least 2 or 3 extra bases after the restriction sites in your primers, the enzymes may not recognize it, hence don't cut... that could be why you're not getting any transformants. Have you tried different vector:insert ratios? Just a question... why do y...

Hello all! I've been trying to transform Agrobacterium tumefaciens, strain EHA105, by the freeze-thaw method. I've done it a few times and didn't get any positive results, even with a liquid nitrogen shock before heating. I was wondering if anyone had problems with this and could give me any tips. P...

Hello!

Can anyone tell/show me how do the several forms of DNA (ds, ss, circular, nicked circular) migrate in an agarose gel? Is there any pattern or it depends on the fragment? A reference would be very welcome.
