Does anyone know how long time (hours - days) it will take if I want to clone a gene in Baculovirus system.

Hi everyone:

I am confused with gene cloning and Gene amplification. Is there a relationship between them?
Why do we have to clone the gene if we can amplify the gene fragment? Is it necessary to amplify the gene after clning for expression?

Hi everyone:

how many enzymes are required to open a cloning vector for insertion of the cloned gene? I am using PFastBac vector and I cut it by one enzyme only. Is this will allow the interested fragment to insert into the vector or I have to use two enzymes???
