NEED IMMIDIATE HELP WITH RNA EXTRACTION!!

Ive been working in a PATH lab for 2 months now. I have been given the assignment of extracting RNA from Formalin fixed Paraffin tissue. So far I have used the RNAeasy Kit for RNA extraction from FFPE and i have also tried a TRIZOL method. NOTHING IS WORKING!!!! I get 0 bands after RT-PCR with B-act...

usually we add proteinease K which denatures all the proteins....if you try adding pro K along with the lyssis buffer chances are you wont get protein interference...... 8)

Has anyone else tried extracting RNA from Formilin fixed parafin tissue? and do you know of an optimal protocol for doing so? because the manufacturers protocol sucks!!!!

How do primers work?

ive been working in a lab for the past month using 3 different primers when i run pcr, but realized that i have no clue, aside that they have "complentary base pairs", on how exactly they break the genome into certain specific genes.

Can someone quickly explain it to me.

yea i do a lot of steps before pcr and it does loose a lot of the DNA , but i think im just gonna go ahead and order new material cause i think your right , i need to start fresh. Thanx a lot for your time and help i appreciate it. ill update u later on how my works goin .

My gel is not the problem and i do use control DNA and i do get bands. I think my main problem is the primers not working correctly. When u make your master mix , how much DNA and primer do u put into each tube? and if for example your using 10 tubes wat is your Master mix consist of? and one last q...
