Hi,I want search some consensus in the genome using blast program on human genome database. But when I enter the sequence which is about 10 base long , the BLAST show me “No significant similarity found” I have enlarge the expect threshold to 10000. nuclear acid probe labeling

I am now preparing run-on assay,and I want some help about it. Dose anyone know it,and tell me some tips about it.

I donnot want label the probe with Isotope(radiolabeled). can anyone tell me the non-isotopel abeling methods . I hope it is sensitive,and popular accepted .

i think you should include a positive control.

how making site-directed mutation on genome DNA

I want to site-directed mutation on genome other than plasmid DNA. Anyone has protocol like this. I donnot want to introduce neo etc antibiotic maker. I need the clear genetic background which the mutation differ to the wild type with only the nucleotide.
