ligation rxn on gel was a smear

i ran a ligation reaction on a gel and got a very faint smear. the smear was so faint i may even be wrong. i loaded 5ul from my ligation which had .1ug dna total. I'm really not sure what happened. I ran a control with everything but t4 ligase and i got two bands. so contaminant can't be in the buff...

How to make diagrams of plasmid constructs?

Do you just use powerpoint? I made a couple mutants and I need to make a diagram of what I did. This is my first time doing anything like this. Any info that would make this easier would be greatly appreciated. Thanks. How do you make arrows (genes) fit around the circle (plasmid)? I can only find s...

Re: Identifying positive bacterial clones - protocol?

this a real newbie question but... if you want to do colony pcr, do you have to streak for single colonies? i can't remember how i was taught a while ago but i thought i just got a new plate divided into about 8 areas and take individual colonies from the original plate and just sort of smear (excus...

Identifying positive bacterial clones - protocol?

I just transformed heat shock cells with the plasmid of interest hopefully with the insert and I grew them on antibiotic plates to select for the plasmid. So now I have maybe 30 colonies and I was wondering what the next step was. Should I take a tiny piece of a number of colonies and streak them on...

'asian flush syndrom' and methanol intoxication

so with asian flush syndrome they have underactive aldehyde dehydrogenase right? and so methanol is converted to formaldehyde and this is toxic when it builds up... therefore people with asian flush syndrome would be more susceptible than someone who metabolizes the aldehyde form fast? or is it not ...

What is NTRI?

I'm reading an article, with no background, on HAART and NRTIs. But I came across NTRI, and I can't find what it is. Can someone explain what it is and even better yet, give me a brief overview of anything related to mitochondrial toxicity of NRTI antiviral drugs.

Need help filling out biology honor society application.

I do not understand these questions fully and I think I'm misinterpreting them. Please help! Here are the questions: 1. What aspects of biological research interest you and why? Is this talking about the different parts involved in research like creating an experiment, obtaining results? or about a ...
