I do not have any purpose of cloning.My aim is to type 300 Campylobacter strains molecularly and look for similarity between isolates; in short my aim is molcular typing. I am using RFLP just for that. Not for cloning or for something genetical method.

My target amplicon is about 1700 bp. After restriction I have difficulty in evaluating the bands sspecially lower than 500 bp.

Do you include positive control for every run and does the positive control give appropriate band? And agarose gel is also important in visualising the results. You should use freshly prepared agarose and ethidium bromide solution.

ELISA protocols changes according to the spesific antigen or the antibody investigated. It also changes according the antigen or antibody whether you use monoclonal or policlonal.

What are you investigating specifically? And for what purpose do you need?
