First Time , Real-Time PCR HELP!!!

Hey, I'm just starting out with this qPCR stuff. I'm looking for for protocols which don't use kits. I can't seem to find any online. I know you use SYBR green as the dye however all the protocols i see use SYBR green kits from Qiagen or Applied biosytems ect. I'm wondering where to get eh SYBR gree...

Degenerate Primers

Hey Brad, Thanks for the Reply. I'm half way there to figuring out my Primers. I tried to use Vector NTI yesterday but like you said it has a high learning curve. I'm currently using a 30-day trail of CLC combined workbench for a Mac which is extremely user friendly and efficient. I've managed to pr...

Degenerate Primers

Hello, I'm very new to molecular biology and I'm helping start up a lab. My supervisor is also new to molecular biology and we are learning as we go so please excuse me if I sound ignorant. I have a few questions about degenerate primers. 1.) Is there an advantage of using the AA sequence over Nucle...

As for the experimental details. Lane 2 and 3 are Identical the only variable I changed was the method of extraction Kit vs Trizol. I have heard that the kit isn't as good at isolating the Small RNa's. I'll eventually be running RT-PCR to determine genes that are differentially expressed during deve...

My Samples are from the leaves of aquatic plants. Each sample was taken from ~7-10mg of tissue. Application site is where the lanes are labelled 1-7. Also the Trizol method shows more faint bands towards the end of the gel. The kit uses glass fiber matrix columns to filter out the RNA. It says it ef...

RNA extraction Kit vs Trizol

Lane 2 and 3 are from the same tissue sample. 7mg of tissue used for each sample. Lane 2 is using a RNA extraction kit using filters and Lane 3 is using the Trizol method. Lane 4 and 5 are also the Kit and lane 6 is Trizol. Do I have DNA contanmination in my Trizol lanes? Why does the kit pick up th...
