Uses for the PCR technique?

... been supplanted by a PCR-like technique where a pair of complementary mutagenic primers is used to amplify the entire plasmid. This generates a nicked, circular DNA which can undergo repair by endogenous bacterial machinery. However, this process does not amplify the DNA exponentially, rather, ...

How do they do it?

... sequences). These enzymes (I think there was 9 or so of them) open up the DNA helix and denature a small region of the DNA, thus making a "nick" on the strand. Next, other enzyme components form the replication fork. Several helicases and topoisomerases are involved here. Only after ...

Re: Identify the nucleic acid!

... further reflection, the question may not be answerable with the information given. I see that lambda exonuclease cannot initiate digestion from a nick or a gap (although it does have very weak activity against ssDNA or so says the descriptive blurb in the NEB catalog), so if it cannot work on ...

One question about Fluorescence in situ Hybridization(FISH)

... in situ Hybridization(FISH) need humannity chamber. Is this instrument necessary? and why? I want to use BAC clone as probe (labelled with DIG-Nick Translation MIX, from Roche) and detected with Fluorescent antibody enhancer set for DIG Detection (also from Roche) . Thanks a lot!!
