Results from our forumPCR & Agarose gel electrophoresis... expose the RNA to light too much? It's possible you denatured them. Also, there may have been RNAses in your extract that could have reduced copy count drastically.

Cell counting question... the thing has an incredibly high error rate anyway (which is why biologists don't really use the thing anymore, there are special devices that can count cells much more accurately).
