GCMS analysis of bacterial cells

Post by stamp608 » Mon Apr 06, 2015 4:59 am

Hi all,
i would like to perform non-targeted GCMS analysis of bacterial cells following treatment with antibacterial compound. i really appreciate if someone can help me on how to prepare the sample prior to GCMS analysis. Please correct my procedure below if i am wrong ya. 10q

a) preparation of bacterial culture
b) centrifugation, to get bacterial cells (pellet)
c) standardization of bacterial density in 1,000,000 CFU/mL (between non-treated control and treated sample)
d) derivatization using relevant solvent (in my case, DMF will be used for alkylation of bacterial proteins)
e) gcms analysis

10q again and nice to discuss with all of you
