NEED HELP!

Hello, everyone. I am puzzled by a question. If I got a gene in
E.coli, how can I know in which operon it locate? And how can I fix
the mechanism by which its trascripition is regulated?

This sounds like a homework question. I'm pretty sure there are easy tests to determine which major operons regulate your gene... Its probably in your book. Changing the mechanism is pretty easy, just change the operator sequence upstream of the gene. I'm not really sure what the limitations of your answer are though, but there are plenty of solutions.

Well if you are in E. coli K12(aka MG1655) or O157:H7 you can lookup the genome. Other wise I suggest:
- Make a probe with your gene
- Southern on PFGE profile or BAC Library
- Sequence clone or cloned PFGE fragment

I am sorry for that I did not describe my question clearly enough.
What puzzled me most is how to find the inducer / inhibitor and other components in a pathway and fix their cascade process.
