Anyone have experience with M63 Minimal Media?

Postby jhard on Fri Jun 01, 2007 2:32 pm

I tried making up a 5x stock solution of M63 and autoclaving it separately from the agar, and had white precipitate. I tried again, autoclaving everything together and the media turned dark (looked almost caramelized).
Anyone know what the media is supposed to look like and the best way to prepare it?

If I remember correctly You are suppose to autoclave separately 3 solution: salts (without MgSO4), Mg SO4 solution and Agar. Then you add the sterile MgSO4 to the sterile salts when they cooled down, and you add agar and water (both sterile) to complete when you need it.

Otherwise, I just realise, filter sterillization is another solution :-)

Thanks :) I may try the sterile filtering if I keep having problems.
