Genetic maps

I'm supposed to write about the basic principles of genetic mapping.
There are so many methods and it confuses me. I really don't know where to start.
Can I have suggestions on what to start with? I can only have 2 pages.

I believe there are 4 strategies for identification and cloning diseasegenes:
- positional cloning
- positional-independent candidate-cloning
- positinal candidate-cloning
- functional cloning

Is it wrong to write about these methods? I believe they are just for locating a mutation that can lead to a disease.

I can suggest papers on positional cloning and functional cloning.

However! I am out of town for the weekend and I don't have my notebooks with me... so i'd have to get back to you on monday. (with suggested papers)

Also, if your paper has to be 2 pages you really can't go into depth too much anyways ... so don't freak-out too much about it if you don't think that you know enough to understand it.... (you'll only be able to give a superficial review with a 2-pager)

If i were to guess 1 and 4 are the classical ways of doing it... and 2 and 3 are "spin-offs" of 1.

anyways... let me know if you want paper suggestions.. . or if you need clarification on 1 or 4 (or a simpler explanation)

positional and funct. cloning are techniques used for genetic mapping.

when i have to identify disease genes i perform a linkage analysis

linkage analysis is one of the first steps... it can give you a good idea of what arm of what chromosome a gene is located at .... once you know that ... you can then see if "your favorite gene" segregates with markers (on the chromosome of the arm that you've already identified)... and once you figure out which gene your gene segregates with... you can use the marker gene as a point to start your chromosome walk (ie. positional cloning).
