Minimal media question

Postby Robin3001 » Wed Nov 05, 2008 1:46 pm

Hi there,

I am doing an honours project using spores of B.subtilis and B.cereus.
I need to prepare a minimal media which does not stimulate germination but supports growth of the vegetative cells. I'm having a really hard time finding out what type of minimal media is suitable for these organisms and my requirements. Someone please help cause i am ready for pulling my hair out!

Thanks,
Robin

I don't get this. You are using spores and you are trying to find a media that does not stimulate germination but supports vegetative cells? How does that work? If they are in spore form that is because they are in unsuitable conditions. So how can some be in vegetative form and some in spore form? Unless some are mutants. Sorry I may be way off, you clearly are in a higher level of studies than I currently am, I just don't get it.

The plan is to see if germination can be stimulated by certain (non nutrient/physiological) chemicals.
Therefore I need to allow for the growth of vegetative cells which have germinated yet i need to rule out the chance of the media alone causing germination.
