Cancer Phenotype Help needed.

Postby keef » Fri Sep 26, 2008 7:32 am

Hi guys. I am having abit of problems with this assignment of mine. Would appreciate if any of you guys could help me a little! thanks!

----------------------
A novel protein, X has been discovered and preliminary experiments indicate that it interacts with p53. Further experiments need to be carried out to validate the observation. Design experiments with illustrations (flow chart) to answer the following:

i a. To show that Protein X interacts with p53
b. To identify the domain of p53 that is important for this interaction?
ii Subsequently, it was seen that the p53 protein with a mutation in the DNA binding domain retains its ability to interact with protein X and induce apoptosis. Explain the observation and design experiments to elucidate the mechanism of p53 - Protein X-mediated apoptosis.

i-I would look for inspiration in the double hybrid system, although it can probably be simplified since p53 interacts with DNA, you just need to define the expected phenotype in case of X/p53 interaction.
