Gel ExtractionHi,
I ran a agarose gel yesterday in TBE buffer. I am just wondering if I leave the DNA band in the gel for two days at room temperature, will the DNA denature? I want to save this gel as a backup for further gel extraction if my sequencing result does not work. Thanks in advance! Maruko
Cut the band, and freeze it. If you store the gel at room temperature, the DNA will likely diffuse in the gel and degradation cannot be excluded. On the other hand freezing the piece of gel might even improve the extraction process.
