i have a gene of interest that has been cloned into a plasmid. but i now need to delete certain segments of that gene & express the protein. is there a protocol available for doing so? i am trying to find any paper which contains such a protocol but am not able to find any. any help will be much appreciated.

I don't know what to suggest without knowing more details. These are internal sequences you need to delete? like you want to remove a specific loop of protein or something like that? Check out Stratagene. They have a range of mutagenesis kits thay may work for you.
