pglo Transformation labI have to write up a conclusion for this lab i have to do for school and i don't understand how I'm suppose to answer these questions. Can someone please help?
The questions are: With regard to lab results, what could you infer by comparing the reults for of the -pGLO LB/Amp plates and the +pGLO LB/Amp plates? With regard to lab results, what could you infer by comparing the results for the -pGLO LB plates and the -pGLO LB/Amp plates? The lab results are: -pGLO LB plate - 335 colonies -pGLO LB/Amp plate - zero colonies +pGLO LB/Amp plate - 37 colonies (that DO NOT glow under UV light) +pGLO LB/Amp/Ara plate - 41 colonies (that DO glow under UV light) please and thank you =) Last edited by konichiwa on Fri Dec 28, 2007 11:20 pm, edited 1 time in total.
Re: pglo Transformation labYou are in fact supposed to think by yourself. Your results are perfectly reasonnable, and should not make any problem in terms of interpretations. I also suggest you to look at a genetic map of pglo and see what genes this plasmid is supposed to contain. Look also if you have information about the promoters of those genes, that would be useful.

Re: pglo Transformation labYou should make a table of what each factor does/tests for. For example the ampicillin plates obviously tests for whether your colonies are resistant.
