problems growing plasmidHi all,
I've been trying to grow up e. coli on an agar plate, and I keep getting small colonies that are very close together. I pipetted 100ul of my transformation onto the plate and then streaked it into 4 quadrants. Are colonies that are the size of approx. a pin head and close together okay? It was difficult to innoculate, but I just wanted to make sure this will work. I did it last week, and my DNA yield after purification was somewhat low. Thanks!
Sounds Ok to me after overnight growth on LB supplemented with anibiotic.
Although after transformations I usually do not streak, but rather spread to get a good separation of colonies Patrick
